Phenyl-substituted dihydropyrazines with DNA strand-breakage activity.

Monophenyl-substituted dihydropyrazines (Ph-DHP-1 to 4) of 2,3-dihydro-5,6-dimethylpyrazine (Me-DHP-1), which have the inductive effects of apoptosis and mutagenesis, were synthesized and their biological effect was investigated in terms of DNA strand-breakage. Differences between the phenyl- and methyl-substituted dihydropyrazines were examined.

Breakage of DNA-membrane complex by bleomycin.

The " DNA-membrane complex" (1200S-400S) obtained by lysis of rat ascites hepatoma cells in a low concentration of sodium dodecyl sarcosinate-sodium deoxycholate sedimented through neutral sucrose gradient more rapidly than the free DNA" (200S) which was released by a high concentration of sodium dodecyl sulfate. The former was decomposed by Pronase in the gradient but the latter was not affect...

The mechanism of DNA breakage by phleomycin in vitro.

Phleomycin induces DNA breakage in vitro in the presence of the sulphydryl compound dithiothreitol. The reaction appears to be free radical-mediated, and requires oxygen and metal ions. Reaction rate is limited by the concentration of oxygen, which is converted to hydrogen peroxide during DNA breakage. However there is no net change in the sulphydryl compound. The proposed reaction mechanism in...

DNA repair: The Nijmegen breakage syndrome protein

The gene mutated in Nijmegen breakage syndrome, a chromosome instability disorder, has been identified and sequenced. The protein product of this gene forms a complex with hMre11 and hRad50--proteins that are involved in repairing double-strand breaks in DNA.

Similarity in the catalysis of DNA breakage and rejoining by type IA and IIA DNA topoisomerases.

Studies of yeast DNA topoisomerase II with various alanine-substitution mutations provide strong biochemical support of a recent hypothesis that the type IA and IIA DNA topoisomerases act similarly in their cleavage and rejoining of DNA. DNA breakage and rejoining by either a type IA or a type IIA enzyme are shown to involve cooperation between a DNA-binding domain containing the active-site ty...
